Explants of Anoectochilus formosanus Hayata were cultured
in vitro in the vessels. Temperature in the culture vessel
was measured with thermocouple and the information was stored
by using CAMPBELL 21X micrologger and single loop controller.
Temperature of the culture room was controlled at 23~25℃.
Straight fluorescent tubes (PHILIPS TLD 83) were employed as
a light source on the culture benches. At 4 hours after the
light period started, temperature inside the vessel reached
27±0.1℃ and dropped to 23±0.1℃ at 4 hours after the dark
period started.
Explants(about 200mg fresh weight) transplanted in vitro on
sugar-free(N) or sugar-containing (S,30g/l) medium which
supplied with banana pulp extract. The culture vessels were
enclosed with medicine papers(P) or aluminum foils(A).
Photoperiodisms of three out of four culture benches , lighted
with PHILIPS TLD 83 tubes were 16hr(light)/8hr(dark)(L),
12hr/8hr(M),
4hr/2hr(S). The last one lighted with PHILIPS Agro-lite tubes and
photoperiodism was 4hr/2hr(Ag).The results of the 1st test showed
that the fresh weight per plantlet of the NA- treatments
were heavier than others after 60 days. Within NA- treatments,
the one with the longest light period has the heaviest fresh
weight per plantlet (560mg) .If the plantlets cultured on the
medium without banana pulp extract, 30g/l sugar-containing medium
showed better results
than sugar-free one.
Plantlets from the culture vessel were transplanted in perlite and
provided with 500ppm Hyponex solution in the second test. Three
levels of both light intensities and CO2 concentrations were applied
to investigate on what is the best combination for growth of plants.
The results showed that 1000 ppm was recomanded for CO2 enrichment
and should be enriched in the light period.The proper light is 54
μmol.m-2s-1. In 134 μmol.m-2s-1 condition , plants showed yellowing
of leaves and some even died. In the 3rd test, the concentration of
CO2 in the culture chamber was controlled within 800~1000 ppm during
the light period and relative humidity was controlled within 85~90%.
All of the light vs. dark period was 16hr vs. 8hr. The medium without
banana pulp extract and agar was replaced by vermiculite. After 60
days, the top fresh weight, root fresh weight and total dry weight
of the NPL treatment was the heaviest out of four treatments (SAL、NAL
、SPL and NPL). The result showed that to enhance the gas exchange
rate of the vessel by using permeable closure ( medicine paper), the
development of explants of A.formosanus can be promoted by CO2
enrichment outside the vessel. Also the sugar-free medium is possible
, if CO2 is enriched and the problem of the containmination can be greatly
reduced.
