Thus, it is possible thathepatopancreas is the extrovarian tissue of Vgsynthesis. The study will provide evidence by invitro culture.Ovary and hepatopancreas from crab at differentovarian stages (oocyte diameter from 167 to 283 um) were incubated in MEM medium containing 14C-leucine.The incorporation of 14C-leucine into tissue wascounted 8 hours after culture. The rate of proteinprotein synthesis in ovary and hepatopancreasdecrease with ovarian maturation. And the rate ofprotein synthesis in hepatopancreas is higher thanthat in ovary. Tissue extracts from 8 hours incubation were analysis by electrophoresis,autoradiography, TCA pe rcipitation andimmunopercipitation. Ovary synthesize three subunitsof Vn. Those proteins that are synthesized byhepatopancreas and can be recognized byanti- vitellin serum, are smaller than the subunitsof Vn/Vg. The molecular weight of the only visiableband in autoradiography is about 85kDa, close to themolecular weight of hemocyanin subunits. The ratesof protein synthesis are 42-199 dpm/mg protein inovary and 544-440 dpm/mg protein in hepatopancreas, both decrease with ovarian maturation. SpecificVn/Vg synthesis was determined byimmunopercipitation with polyclonal Vn-specificantiserum and protein A. The percentage of Vn/Vgspecific synthesis in the total protein synthesisare 3.7-11.0% in ovary and 2.4-11.0% inhepatopancreas. Judging the volume of the twoorgans, Vn/Vg synthesis in hepatopancreas are muchhigher than in ovary during those ovarian stagesstudied.The results led to the following conclusions: 1).Both ovary and hepatopancreas are the sites of Vnsynthesis in Scylla serrata. 2). Hepatopancreas isthe major source of Vn/Vg, especially in the earlystage of ovarian maturation. 3). The rate of proteinsynthesis in ovary and hepatopancreas decrease withovarian maturation. 4).
