This study employed RAPD analysis using total DNA of soybean as templates. The genetic similarity coefficients were calculated and then grouped by cluster analysis to investigate the relationship among intra- and inter- species. The main results were summarized as follows: 1.The similarity coefficients of 38 local lines were from 0.53 to 0.98. It was indicated that the differentiation among varieties had been occurred. However, it was found that there were 30 varieties whose similarity coefficients were greater than 0.8. It meant that the main variation was caused by few varieties. Based on the results, thirty-eight varieties could be clustered into 26 germplasms for conservation. 2.Of the subgenus Soja, the kinship of semi-wild species G. gracilis was between G. max and G. soja, and could be grouped as the same cluster with G. max. Although G.gracilis had a close relationship with G. max, we still could find some specific bands for G. gracilis, and these bands could be use as molecular markers. In regard to the two wild species of subgenus Soja, G. soja got closer relationship with G. max than with G. formosana. However, G. formosana had the smallest intraspecific variation. And it appeared a significant difference from other species on RAPD profile. In this study, specific bands for G. formosana were also observed. 3.G. dolichocarpa showed significant differences in comparism with G. tomentella and G. tabacina in DNA sequences. The name G. dolichocarpa was appropriate. The kinship of G. dolichocarpa was between G. tomentella and G. tabacina, but should be closer with G. tomentella. 4.G. tomentella in subgenus Glycine had comparatively close kinships with the lines in subgenus Soja. Among these, G. formosana in subgenus Soja showed a closer kinship with G. tomentella than others. However, the similarity coefficient of subgenus Glycine and subgenus Soja was only 0.12, and the kinship was very far. Thus, G. tomentella could not be proned as the ancestor of subgenus Soja. 5.RAPD had a nearly equivalent performance with RFLP in constructing the kinship among interspecies, so it was a fast and efficient method in application.
