Summary:碩士 === 中國醫藥學院 === 中國藥學研究所 === 85 === In the present study, we investigated the sedative, analgesic, anticonvulsive, antipyretic and hypothermic effects of the various preparations of Paeoniae Alba Radix (PAR, PARa, PARb and PARc) and Shao-Yao-Kan-Tsao-Tang (SKT, SKTa, SKTb and SKTc) by pentobarbital-induced hypnosis, acetic acid-induced writhing response, formalin test and LPS-induced fever. We also detected the content of the major active constituents (paeoniflorin, paeonol ) of the various Paeoniae alba radix and Shao-Yao-Kan- Tsao-Tang.The various preparations of Paeoniae Alba Radix and Shao-Yao-Kan-Tsao-Tang prolonged the sleeping time induced by pentobarbital. PAR, PARa, PARb and the various preparations of Shao-Yao-Kan-Tsao-Tang inhibited the writhing response induced by acetic acid. PARa, PARb, SKT, SKTa and SKTb inhibited the early phase produced by formalin. PARa, PARb and the various preparations of SKT inhibited the late phase produced by formalin. The various preparations of Paeoniae Alba Radix, SKT, SKTa and SKTb had hypothermic effect in the normal rats. PARa, PARb, PARc, SKTa and SKTb inhibited the hyperthermia induced by LPS. PARa, PARb, SKTa and SKTb prolonged the initiation of clonic convulsion produced by strychnine.In the HPLC analysis, the paeoniflorin and paeonol content in PARa, PARb, SKTa and SKTb were higher than PAR, PARc, SKT and SKTc. Furthermore, the paeoniflorin and paeonol content in the Shao-Yao-Kan-Tsao-Tang was higher than those in Paeoniae Alba Radix.In conclusion, the alcohol and acetic acid preparations of Paeoniae Alba Radix and Shao-Yao-Kan-Tsao-Tang posessed the better sedative, analgesic, anticonvulsive, hypothermic and antipyretic effects than the other preparations. They also contained the higher contents of the major active constituents. Paeoniflorin and paeonol could be the active constituents in Paeoniae Alba Radix and Shao-Yao-Kan- Tsao-Tang.
