It was found that high concentration of glycerol ( 6% , 20% ) andNaCl ( 3% , 10% ) could stimulate the proline productivity of C. glutamicum.The proline production in the media containing 8% NaCl was 8 time more thanthose in the regular media. To optimize the medium composition, responsesurface methodology ( RSM ) with three- variable ( X1=ammonium chloride ,X2=NaCl , X3=glucose ) and five-level central composite design was adopted.It was shown that the optimal concentration of ammonium chloride , NaCl , and glucose were 1.4 , 6.9 , 1.76% , respectively. On this condition , prolineproduction of 72-hour fermentation was 0.1274 umole/ml , and the predictiveproduction was 0.1233 umole/ml. The difference is insignificant according tot-test (p > 0.05 ). The optimum medium formila was also applied to 2-liter batch fermentation in a jar fermenor. The production of proline after 72-hourfermentation was 0.10 umole/ml, approximately 78% of the production byshaking flask fermentation.
