Five healthy dogs of body weight ranging 23-33 kg were hemodialyzed with heparin as the sole anti- coagulant which was administered at a dose of 40 IU/kg (initial dose) and 30 IU/kg/day (maintenance dose). Groups II and III were hemodialyzed with heparin at the same dose and together with two regimens of ticlopidine, 50 mg/kg/day and 100 mg/kg/ day respec- tively. Ticlopidine was administered orally once a day at the doses mentioned above 72 hours prior to hemodialysis, and the evaluation of ALT, BUN, creatinine, blood cell count, APTT and fibrinogen levels was carried out before each administration. The period of each hemodialysis was 3 hours. Parameters monitored during hemodialysis included APTT, fibrinogen levels, blood cell count, BUN, creatinine and residual blood weight. Systolic arterial pressure, diastolic pressure, pulses and mean arotic pressure were also recorded via automatic sphygnooscillometer at five minutes intervals before, during and after hemodialysis. No significant differences of those evaluated parameters mentioned above were found among dogs which were pretreated with ticlopi- dine. A significantly lower degree of hemodialysis-associated leukocytopenia was found in both groups II and III than that of group I. However, no significant differences were observed between the groups II and III. In addition, dose-dependent reduction of residual blood weight was observed in the groups II and III. A much less degree of hemodialysis-associated thrombocy- topenia was observed in group III as compared to group I. According to the salutary effects demonstrated above, the combi- nation of heparin and higher ticlopidine regimen may be more clinically benefic.
