proteins during oogenesis. According to the sizes, five groups
of carp oocytes were classified:perinucleolus stage, oil
droplet- yolk vesicle stage, primary yolk stage, secondary yolk
stage and tertiary yolk stage. Total RNA was first extracted
from carp oocytes in order to quantify the ZP2、ZP3 and fibroin-
like protein mRNA by Northern blot analysis. Secondly, the PBS-
insoluble fraction was prepared from carp oocytes to quantify
the protein content of ZP2、ZP3 and fibroin-like protein by
immunodot blot. The mRNA content of ZP2 and ZP3 decreased
rapidly when oocytes developed from the perinucleolus stage to
the primary yolk stage, but changed slightly during the
development from the primary yolk stage to the tertiary yolk
stage.The amount of ZP2 and ZP3 mRNA dropped from 1.0 ng and
1.5 ng in one oocyte of the perinucleolus stage to 0.2 ng and
0.2 ng in one oocyte of the primary yolk stage, respectively.
The mRNA content of fibroin -like protein also decreased
markedly from 1.6 ng in one oocyte of the perinucleolus stage
to 0.3 ng in one oocyte of the secondary yolk stage.
Thereafter, the fibroin-like protein mRNA content raised when
oocytes developed to tertiary yolk stage. On the other hand,
the protein content of ZP2 and ZP3 increased slowly when
oocytes developed from the perinucleolus stage to the primary
yolk stage but increased 180 fold and 279 fold during the later
stages of oogenesis, respectively. The fibroin-like protein
increased significantly (about 53 fold) until the development
from the secondary to the tertiary yolk stage.In summary, the
transcription of carp ZP2、ZP3 and fibroin-like protein genes
occurs in the early phase of oogenesis and their transcripts
are translated into egg membrane proteins until vitellogenesis.
