HL-60 cells revealed apoptosis when terminally differentiated (Solary et al, 1994). This thesis investigated if TPA might induce differentiation and apoptosis in human cervical cancer SiHa cells. After treate for 3-4 days, SiHa cells revealed decreased proliferation rate, G1 stasis and decreased expression of K-19 which indicated TPA induced differentiation of the cells. In SiHa cells treated for 7-9 days, chromatin condensations were rvealed by DAPI stain and electron microscopic observation, and DNA fragmentation was found on agarose gel electrophoresis. These results suggested that TPA induced apoptosis in SiHa cells. In addition, autophagosomes of mitochondria at various stages were found in the differentiating SiHa cells. Since autophagocytosis of mitochondria was shown to play an important role in differentiation of Leydig cells (Tangi,1988), we proposed that autophagocytosis of mitochondria might be one of important mechanisms involved in TPA-induced differentiation of SiHa cells.
