Summary:碩士 === 中山醫學院 === 生物化學研究所 === 84 === Immunofluorescece analysis using a variety of human autoimmune sera showed a typical dot-like nuclear staining,called nuclear domain(ND)10,also desc-ribed as POD or Kr bodies,suggesting that this structure consists of seve-ral proteins.Two of the ND10 proteins,Sp100 and PML are genetically chara- cterized and show homology with several transcription factors; the first a-utoantigen predominant in patients with primary biliary cirrhosis(PBC) and the second transformation and cell growth suppressing protein aberrantly expressed in promyelocytic leukemia cells.In this study,we found a sarcoi- dosis patient whose serum recognized two proteins with Mr of 92 kDa and 87 kDa in HeLa cell and lymphocyte cell lysate.The serum also recognized two proteins with Mr of 70 kDa and 66 kDa in rat liver cell nuclear extract.W- hen 60-100% ammonium sulfate was added to rat liver cell nuclear extract,a protein with Mr of 58 kDa was recognized by this patient's serum.By the u- sing of DE-52 cellulose,phenyl-agarose and Superdex HR200 chromatography a protein with Mr of 58 kDa which againsted the serum of sarcoidosis patient was purified from rat liver cell nuclear extract.To determine the NH2-amino acids sequence of this 58 kDa protein is doing now.
