In general,high gelstrength and lower yield of agar product was obtained when dried Gracilaria tenuistipitata was soaked for two hours before alkaline treatment. Sulphate eliminase, the enzyme existing in Gracilaria tenuistipitata, affects the gel strength of agar, however, depolymerase of Gracilaria tenuistipitata affects the yield of agar extraction. The optimal pH value of A and B sulphate eliminases is 8.2 and C is 7.5. The optimal temperature is 37℃. The molecular weight of A, B and C were all 140,000. The molecular weight of The optimal pH value of agar depolymerase was 6.5. The 10mM Mg2+ solution, Ca2+ and Zn2+ active agar depolymerase,however,more than 20 mM Mg2+, Ca2+, Co2+, Cd2+, Mn2+, Ni2+ and Fe3+ result in Consequently, the results of this study showed that adjusting pH value of socking water up to 7.6 and lasting for 2 hrs and simul- taneous mixing 20mM Mg2+ for activating sulphate eliminase was
