squamous epithelia express 10 to 30 persents of their
total protein as keratin. The two types of keratins are
specifically expressed in these cells. They are the type I
keratin K5 with a molecular weight of 58 kelodalatons and its
corresponding partner , type II keratin K14 with a molecular
weight of 50 kelodalatons. The mechanisms underlying the
regulation of keratins is an important step in understanding
the epidermal differentition. In order to delineate the
regulatory mechanism of K14 gene, the pK14CAT(-274) and pK14
CAT(-2000) were used in this investigation. The experssion of
choloramphenicol acetyl transferase of the two constructs were
analyzed in primary culture and cell lines, which included
epithelial and non-epithelial cells. The results
indicated that the pK14CAT(-274) and pK14CAT(-2000)
were functional in all epithelial cells but not in non-
epithelial cells except that in smooth muscle cells.
Interestingly, those two constructs were both very active
in stratified squamous epithelial cells than that in
simple epithelial cells. In comparison, the regulatory
expression of the pK14CAT(-274) in stratified squamous
epithelial cells was stronger than the pK14CAT(-2000).
When CAT-plasmid containing host cells treated with TPA for
48 hours, the significant down expression of the pK14
CAT(-2000) can be observed. In conclusion, the pK14CAT were
specifically expressed in epithelial cell and the pK14
CAT(-2000) were specifically expressed in stratified
epithelial cells.
