The genotype and predicted transmitting ability of dairy bulls were compared, and using random amplified polymorphic DNA polymerase chain reaction (RAPD PCR) to establish the DNA fingerprints of dairy bulls. Kappa-casein genotypes of 43 dairy bulls were analyzed using PCR with restriction fragment length polymorphisms (PCR- RPLPs), and the genotypes identified were 35 AA, 7 BB, and 1 BB. The B gene frequency was found to be 10.5%. Comparison between genotypes and transmitting trait evaluation indicated that bulls with AB type gene have higher predited transmitting ability for the yield of milk, protein and fat than those with BB type gene. In the alpha-lactalbumin genotyping of 45 dairy bulls, the genotypes identified were 2 AA, 18 AB, and 25 BB. The A gene frequency was found to be 24.4%. Comparison between genotypes and transmitting trait evaluation indicated that bulls with AA type and AB type gene have higher predicted transmitting ability. DNA fingerprints were established by RAPD PCR. By screening with 140 primers, 16 fingerprints and 43 stable amplified products were obtained. The stability of the fingerprints was confirmed by repeating the tests for individual and pooled semen DNA. The results of this study can serve as a reference for dairymen to select the frozen semens of dairy bulls. The establishment of DNA fingerprints may also contribute to the improveement in the selection of dairy cattle. Frank Chiahung Mao 毛嘉洪先生 1995 學位論文 ; thesis 97 zh-TW
