We have developed a convenient High Performance Liquid Chromatogr- aphy (HPLC) assay employing fluorescent detection for the accu- rate determination of human plasma or microdialysis perfusate into methanolic monobromobimane, which accomplished protein pr- ecipitation and thiol derivatization simultaneously. Thorough deproteinization of the tissue homogenates and whole blood were achieved by heating the sample followed by addition to methano- lic monobromobimane. Separations of derivatized materials were achieved on either conventional or narrow-bore columns. Major results from applications of the established assays wewe:(1) glutathione levels in plasma and whole blood decreased with increasing ages in normal Chinese;(2) glutathione leves in brain and spleen decreased with increasing ages in Wistar Kyoto rats; (3) after concentric exercises,plasma oxidative stress increased in patients with coronary artery diseases;(4) glutathione levels were significantly higher in human oral tumor tissues than those in normal tissues ;(5) L-buthionine sulfoximine and diethyl maleate had the same effect on lowering GSH levels in various rat organs such as liver , heart and spleen ;(6) brain extracellular glutathione levels rapidly increased after cerebral ischemia in rats ;(7) elevation of glutathion levels in plasma after alcohol consumption is probably due to the release from erythrocytes rather than that from liver.
