Summary:碩士 === 國立臺灣大學 === 獸醫學系 === 82 === To develop a sensitive diagostic reagent against infection of mouse hepatitis virus (MHV), one of the most commonly occurring pathogen in laboratory mouse colonies, hybridoma cell lines producing monoclonal antibodies to a Taiwanese strain of MHV were established. Liver emulsion of this local isolated MHV was used as immunogen to infect 1 to 3 day old mice intraperitoneally. By indirect immunofluoresence (IF), monoclonal antibodies reacted in the cytoplasma of infected cells were identified. Among the 62 IF postive clone, antigenic specificity were further recognized by radioimmunoprecipitation and three kinds of viral structral protein, spike (31 clones), matrix (3 clones) and nucleocapsid (8 clones), were confirmed. All these antibodies belong to various IgG subclasses, e.g., IgG1, IgG2a, IgG2b, as determined by ELISA. Plaque reduction tests were done and 4 clones shown positive to neutralize MHV Taiwanese strain.
