PSP was induced in female hamsters by mating with vasectomized male hamsters at estrus.Decidualization of one uterine horn in response to artificial stimuli was induced in hamster on the fourth day of PSP. Hamsters were decapitated at 3:00 p.m. and 7:00 p.m. on the fourth day, and at 9:00 a.m. to 10:00 a.m. from the fifth to the tenth day of PSP. The mesometrial(MS) and antimesometrial(AMS) deciduoma were separated and cell suspensions were obtained following trypsin and collagenase treatment. The incorporation of BrdU into replicating DNA was monitored as a parameter for DNA synthesis and was measured using a monoclonal antibody against BrdU and EIA. The cell bioactivity was detected using the MTT colormetric assay. Uterine weight rose after the induction of decidualization and reached the maximum, which was 15 times of control, on the ninth day of PSP. The results showed that there were different developmental patterns of DNA synthesis and cell bioactivity during decidualization in PSP hamster. The deciduomal DNA synthesis activity was elevated sharply 10 hr after stimulation; and the maximum activity was reached on the fifth day of PSP. The DNA synthesis activity declined gradually thereafter. The deciduomal cell bioactivity did not change at the early stage of decidualization. It remained low till the ninth day of PSP. No difference of DNA synthesis activity and cell bioactivity were detected between the MS and AMS deciduoma.
