spelling ndltd-TW-082NCHU05410112015-10-13T15:33:32Z http://ndltd.ncl.edu.tw/handle/52837243140235878823 Studies on the Antibacterial Activities of Quinolones against Edwardsiella tarda Isolated from Cultured Eels in vitro Quinolone類藥物對Edwardsiellatarda之抗菌效力研究 Deng;Ching-Ying 鄧晶瑩 碩士 國立中興大學 獸醫學系 82 1992-1993年間由中部養殖鰻所分離到 Edwardsiella tarda 菌株共有90 株(分離率45.8%),測定nalidixic acid (NA) 、flumequine (Fq) 、 oxolinic acid (OA) 、norfloxacin (NF) 、enrofloxacin (EF) 、 chloramphenicol (CM) 、oxytetracycline (OTC) 、streptomycin(SM) 等抗菌劑對其之最低抑菌濃度 (MIC) 。結果顯示E. tarda對 enro- floxacin 、norfloxacin感受性最高(MIC50 均為 0.125mcg/mL); oxolinic acid (OA) 、flumequine (Fq)次之 (MIC50 均為1 mcg/mL); chloramphenical (CM)更次之(MIC50 則為 4 mcg/ml):nalidixic acid (NA)及streptomycin(SM)較差(MIC50均為16 mcg/ml); oxytetracycline (OTC) 最差(MIC50為128 mcg/ml)。 由MIC值之測定 結果顯示這些菌株有許多已具有抗藥性:對常用之抗菌劑如 oxytetracycline (OTC)具有抗藥性菌株之百分比高達 83.3% ;對 streptomycin (SM)之抗藥性菌株之百分比則為45.6% ;對CM及NA具有抗 藥性之菌株之百分比均為 37.7% ;對新型 fluoroquinolone藥物則無抗 藥性菌株產生。由測定五種quinolone類藥物間之交叉抗藥性試驗結果得 知,NA、Fq、OA、NF及EF五種抗菌劑之間具有不完全交叉抗藥性反應存在 。並且比較1993年之E. tarda分離株與1992年前之分離株對藥物之感受性 試驗,結果顯示1993年所分離之菌株對OTC之抗藥性之百分比,較1992年 前之分離株有顯著性的增加( p<0.05 )。比較測試4-quinolone類藥物 及CM分別於1倍、5 倍、10倍及20倍MIC劑量時藥物對E. tarda菌之殺菌效 力。結果顯示以EF最具殺菌活性,10倍MIC值劑量時,於30分鐘即有99%以 上之殺菌能力,3小時可達100%之殺菌效力;於20倍MIC值劑量,則於2小 時有100%殺菌力。NF之殺菌能力僅次於EF,並且其殺菌力與EF相似,但無 EF之快速。OA及FQ殺菌效力相同;NA及CM則最差。溫度對藥物活性之影響 結果顯示:在第一個小時時以低溫 20℃)時之抑菌效力較高溫(30℃) 時為佳,然於第2個小時後溫度對藥物之抑菌力則無影響。酸鹼值對細菌 的藥物感受性影響試驗結果顯示,pH6.0時藥物之制菌能力較差,MIC值可 增加64倍以上。而pH 9.0 時藥物之MIC值反有降低之情形;即表示藥物抗 菌活性較好。在離子濃度對藥物抗菌效力之影響試驗結果顯示;在含8 mM 鈣離子時其MIC值會增加16∼32倍;但在含32 mM 鎂離子時MIC值卻可增 加128∼256倍;即藥物會隨著離子濃度增加而使藥物抗菌活性亦降低。 The aims of this study were to detect the effects of anti- bacterial activities of several antimicrobics including enro- floxacin(EF),norfloxacin(NF), oxolinic acid(OA), flumequine( Fq), nalidixic acid(NA), streptomycin(SM), chloramphenicol(CM), and oxytetracycline(OTC) against Edwardsiella tarda isolated from cultured eels by temperature, pH, and divalent cations in vitro. The results of sensivity test indicated quinolones had better antibacterial activities than other antibiotics. There were incompletely cross resistant relationship among quinolones. The data of effects tests suggested quinolones had the best bacterial inhibition at low temperature, high pH value, and low divalent cations concentrations.
