Summary:碩士 === 國立中興大學 === 獸醫學系 === 82 === The aims of this study are to evaluate the efficacies of 11 gl- ycans(Bar, curdlan, dextran sulfate, inulin, krestin, laminar- an, levan, PO, scleroglucan, yeast glucan, and zymosan) to pr- otect four different species of fish, bighead carp( Aristichthys nobilis), grass carp (Ctenopharyngodon idellus), milk fish (Cha- nos chanos), and tilapia (Tilapia aureas P.) against bacterial infections in vivo. The result revealed that several glycans, Bar, krestin, sclerog- lucan, and zymosan, did significantly protect the four kinds of fish against the bacterial infections. At the same time, the data shown that the antibacterial mechanism of glycans associat- ed with the activation of the nonspecific phagocytosis. The results of challenging with Aeromonas hydrophila study sugg- ested that four glycans, Bar, krestin, scleroglucan, and zymosa- n, significantly increased the survival rates in grass carp(6 0﹪, 70﹪, 90﹪, and 60﹪) and tilapia(80﹪, 60﹪, 70﹪, and 60﹪ five glycans, Bar, curdlan, krestin,scleroglucan,and zymos- an,did significantly increase the survival rates in both bighead carp(70﹪, 40﹪, 40﹪, 70﹪, and 50﹪) and milk fish (50﹪, , 40﹪, 60﹪, and 40﹪). The results of challenging with Edwardsiella tarda study indicated that four glycans, Bar, krestin, scleroglucan, and zymosan, did significantly raise the survival rates of all fishes. Studing on the kinetics of the numbers of nitroblue tetrazolium (NBT)- positive staining cells by the Adherence/NBT Assay eva- luates the non-specific immunity responses in the fish injected with the five glycans including Bar, curdlan, krestin, sclerogl- ucan, and zymosan, by the dose of 10mg/kg bw, respectively. The result revealed that those five glycans, except curdlan in milk fish, did significantly increase the numbers of NBT- positive staining cells in the four species of fish.
