''Samantha'',''Landora'' and ''Pitica'' roses were studiedin vitro.
The level of sucrose in medium was at 3.8、7.5、15 or 30g/L.The
more growth of ''Samantha'' and ''Landora'' explants was cultured
on the medium contained higher concentration of sucrose.
However, the best growth of ''Pitica'' explants was cultures on
the medium contained 15g/L sucrose. And the multiplication rate
of shoot explants increased as the sucrose concentration
increased in medium.The CO2 concentration in vessel was higher
when the sucrose concentration of culturing medium was higher.
The growth of ''Samantha''and ''Landora''explants cultured under
high PPF(70.mu./m2/sec) was greater than that cultured under
low PPF (35.mu.mol/m2/sec).Moreover,under high PPF,the growth
of explants cultured on a medium contained 15g/L sucrose was
greater than that cultured on a medium contained 30g/L sucrose.
Under high PPF,''Landora'' explants did not survive on a medium
without sucrose but 40% of ''Samantha''explants survived.After
cultured under high PPF for 4 weeks, the lowest CO2
concentration in vessel was in which the explants grow on a
medium contained 15g/L sucrose. The dry matter of explants was
higher and the leaves was dark green with CO2 enrichment.
Explants cultured on a medium contained 7.5g/L sucrose and
enriched with CO2 was the vigorest and a medium contained 15g/L
sucrose enriched with CO2, the leaves of ''Samantha'' explants
was distortion and the low leaves of ''Landora'' explants was
yellowing.Under high PPF, on the leaf of explants there were
ellipsoid stomata with different aperture. Furthermore, the
leaf had typical palisade mesophyll and there wrer starch
grains in chloroplast. The additional CO2 enrichment culture,
the aperture of stomata was smaller or closed, and the leaf
thickness was greater and there was the biggest starch grain if
the explants cultured on a medium contained 7.5 g/L.
