Summary:碩士 === 中國醫藥學院 === 中國藥學系 === 82 === Palmatine, an active constituent isolated from Chinese herbs, such as Fibraura recisa, Cordalis yanhusuo,Coptis chinensis, Phellodendron amurense et al, possesses anti-bacterial, sedative and antiarrhytymic effects. In this study, the pharmacokinetic and bioavailability of palmatine in rabbit were evaluated. A selective and sensitive reversed-phase high- performance liquid chromatographic method were developed for the determination of the palmatine concentration in plasma. The separation method is effective and precise. It could be applied in the determination of the palmatine concentration in plasma . The bioavailability of the palmatine were studied in 6 rabbits by a three-way cross-over design. The venous blood samples were collected at several fixed time intervals after the drug administration, for I.V. (10 mg/kg), I.M. (10 mg/kg) and P.O. (90 mg/kg), respectively. All plasma samples were assayed by HPLC method and data were treated by computer programs: LAGRAN- P and PCNONLIN.When the palmatine give to the rabbit by intravenously, it exhibits two compartment pharmacokinetics, but if give orally, it exhibits one compartment pharmacokinetics. The pharmacokinetic behaviors following these three different route of drug administration are quit different. The bioavailability of I.M. rout was approximate to 100%. However, the bioavailability of P.O. was extremely low, only 1.7%.
