The root of Aconitum yamatoanum has been used as one ofthe most important Chinese drugs(Fu-tzu)prescribed together with other herbs,as an analgesic in the treatment of rheumatism and neuralgea. The roots collected from plants naturally grown in the mountainous areas andpropagated by tra- ditional methods are far from enough for medicinal use. The tissue culture method of mass propagation was therefore studied. The results are summarized as follows. 1. The germination rate as well as the growth of the seedlings of A. yamatoanum were superior in MS medium containing 1 mg/l 2,4-D and 1 mg/l BA. 4. The callus induction rate from leaf blade was higher than that from petiole and root. The callus originated from leaf blade also tended to become browning more easily than those of other origins. 6. Callus induced from the leaf blade possessed the ability of root differentiation. While cultured in medium containing MS salts , 5 mg/l BA ,0.5 mg/l NAA ,and 5 mg/l AgNO3 ,callus from the differentiated root could produce buds under light condition. 9. Adequate browning of the leaf blade kept under light condition was beneficial to the successful production of adventitious bud in vitro. 12. The best rooting medium was 1/2 strength of MS salts, 5﹪ sucrose and 1 mg/l NAA. The highest rooting percentage of 70 ﹪ was recorded. 13.
