Description
Summary:碩士 === 國立臺灣大學 === 食品科技研究所 === 81 === Bilirubin from pig bile was analyzed in this experiment. The bilirubin diazo products, obtained from diazo reaction using diazotized o-ethyl anthranilate as the diazo reagent, were analy- zed by UV-VIS and high performance liquid chromatography. After diazotization, the diazo products were also separated with column chromatography and the main fractions furthermore underwent tran- sesterifica-tion and acetylation. NMR and Mass spectra of these reaction products were used as the corroborative evidence for the previous analysis. Results of these investigations showed that there were probably three types of bilirubin existed in pig bile ,namely, alpha, beta, and gamma forms. The phenol-sulfuric acid method was used to measure the total amount of sugar released from bilirubin after transesterification. The quantitation showed that one molecule of bilirubin, on the average, was combined with about 1.49 molecule of glycosyl group. It was also found that the amount of bilirubin in pig bile was different among pigs; about 12 to 23 miligram/100 mililitre.
