We examined 28 samples from 24 patients with AML, the MDR1 RNA levels were determined by slot blot analysis and compared with those of drug-resistive and multidrug resistant cell lines. The MDR1 RNA expression was negative in 28.6% and positive in 71.4% of the patients. Age, WBC count, percentage oflast, LDH and induction chemotherapy were not significantly difference between MDR1 RNA-negative and positive patients. However, the complete remission rate was lower for patients with MDR1 RNA positive than for those negative group [26.3% (6/19) vs 80%(4/5)]. We had three cases, their MDR1 RNA expression progressively elevated by disease progression and number of chemotherapy. So we suggest that high frequency of MDR1 gene expression in the AML, and the elevated MDR1 RNAevels in the AML were associated with lower complete remission rate, resistant disease, early death, and shorter disease-free survival. So that the expression of MDR1 is a poor prognostic factors for the AML treatment, and it may play an important role in resistance to chemotherapy.
