Proliferating cell nuclear antigen (PCNA) , also known as cyclin or the auxiliary protein of polymerase δ, is a growth - regulated nuclear protein. One of the rat genomic DNA fragments containing PCNA coding regions has been cloned and partially sequenced. This clone was 14 kb in Iength and contained the full - length of PCNA gane in its 8 kb EcoRI fragment. Sequence analysis of a 1.6 kb fragment which covered the exon 1 and its 5' and 3' flanking regions revealed features and structural motifs which were also found in the respective areas of human and mouse PCNA genes. Among these, absence of TATA box and the presence of several SPI sites were noted, a typical characteristic of many house - keeping genes. A preliminary test of promoter activity for the 1.6 kb fragment has been conducted. The transient expression of chloramphenicol acetyltransferase in CHO - K1 cells transfected with the concerned DNA was used to measure the strength of the promoter. The positive results of this test were in consistency with the presumption that 1.6 kb fragment contained the putative promoter of the rat PCNA gene.
