Summary: | 碩士 === 國立臺灣大學 === 農業化學研究所 === 65 === In this study, the TNBS procedure proposed by Kakable and Liener was modified and used to determine the available lysine of several food and feed samples, and the results obtained were compared with those obtained by using FDNB method and the Kakade and liener's original TNBS method.
The ratio between sample weight and acid volume used for hydrolyzing the trinitrophenylated samples may affect the observed values of available lysine. In the presence of starch or lactose, lower values of □ TNP - lysine contents were obtained, and the addition of picric acid reduced this effect to a certain extent, although not completely. These results are similar to those observed by using FDNB reagent for the determination of available lysine.
The feasibility and precaution of using free lysine an an internal standard to check the recovery of □ - TNP - lysine during acid hydrolysis of the trinitrophenylated samples were also discussed. The effect of the degree of partial hydrolysis on the observed values of □ - TNP - lysine contents in trinitrophenylated proteins was recognized in this study, although its effect on the determination of available lysine of food and feed samples was not significant.
