Epidemiological evidence has conclusively demonstrated that chronic arsenic exposure is associated with an increased risk of skin and several internal human cancers, including lung, kidney, colon and bladder cancers. Yet the mechanism by which arsenic acts to induce carcinogenesis has not been established. Unlike most chemical carcinogens, arsenic is inactive as a point mutagen; however, it has been shown to induce chromosomal aberrations. The lack of arsenic mutagenicity has led to two hypotheses predicting a possible role for arsenic in the process of carcinogenesis. One hypothesis predicts that arsenic may induce amplification of genes important in the progression of cancers. Alternatively, it has been proposed that arsenic acts as a co-carcinogen, working synergistically with other environmental mutagens to induce cancers. In an effort to explore these two hypotheses, I examined the effects of arsenic as a mutagen or co-mutagen with UV-radiation on the induction of gene amplification in the yeast model, Saccharomyces cerevisiae. I report a ~30-fold increase in ADH4:CUP1 amplifications in yeast cells exposed to arsenic. Treatment of cells with arsenic in combination with UV-radiation showed no enhancement of ADH4:CUP1 amplifications above exposure to arsenic or UV alone. These results indicate that arsenic may act alone to generate gene amplifications as its role in inducing human carcinogenesis. To obtain additional information about the mechanism of arsenic-induced amplification, the structural nature of the amplified DNA was analyzed.
