Biochemistry Biology Biomedical Research Cellular Biology Medicine Oncology RRD2 RRD4 cancer platinum drugs ovarian cancer chemotherapy apoptosis This thesis investigates the mechanism of action of two novel anticancer drugs, RRD2 and RRD4. RRD2 and RRD4 are platinum analogs that show promise in the treatment of ovarian cancer1. Little is known about the mechanism by which these two drugs induce apoptosis in cancerous cells. This thesis examines the effects of RRD2 and RRD4 on the expression and activity of various proteins in an effort to understand the changes that result in drug-induced apoptosis. Alterations in the expression of intrinsic and extrinsic apoptotic pathway proteins, insulin-like growth factor signaling (IGF) proteins, heat shock proteins, cell cycle regulatory proteins, and phosphatidylinositol-3 kinase (PI3K) signaling proteins are investigated. The effects of RRD2 and RRD4 treatment on acid sphingomyelinase activity are also elucidated. Experimental results suggest that RRD2 and RRD4 induce apoptosis via the intrinsic and extrinsic apoptotic pathways. Results also demonstrate pro-apoptotic alterations in the expression of cell cycle regulatory proteins and modifications of the IGF and PI3K signaling pathways. Further, RRD2 and RRD4 cause robust activation of the acid sphingomyelinase enzyme.
