Biomedical Engineering E-selectin Galectin-1 Breast Cancer Mac-2BP Invasion of other tissues during bloodborne metastasis requires the adhesion of cancer cells to the endothelium by specific fluid shear-dependent receptor-ligand interactions. It is hypothesized that ligands are shared between the endothelial adhesion molecule known as E-selectin and Galectin-1 (Gal-1), both of which are upregulated during inflammation and cancer. Flow chamber analysis of ZR-75-1 breast cancer monolayers and dynamic biochemical tissue analysis (DBTA) of mucinous breast carcinoma revealed a shear-dependent binding interaction between Gal-1hFc treated surfaces and E-selectin coated Protein A beads causing 300% and 142% binding increase of the beads, respectively. Immunocyto- and immunohistochemical analyses showed that Gal-1 and E-selectin fluorescent signals on ZR-75-1 cells and mucinous carcinoma tissues colocalized at approximately 75% for each assay. Immunoprecipitation of Mac-2BP from breast cancer whole cell lysates displayed that Gal-1 and E-selectin share Mac-2BP as a ligand. Finally, widefield FRET analysis of E-selectin and Gal-1 stained ZR-75-1 cells indicated that previous colocalization data may not be due to Gal-1 and E-selectin on the same ligand.
