Biochemistry Molecular Biology UVB calpain calpeptin IkBa CK2 skin cancer Skin cancer is the most prevalent cancer in the United States and is usually induced by UV exposure. UV radiation is known to activate both calpains and NF-¿¿¿¿B, two proteins whose activity has been implicated in cancer development and progression. The inhibitor of ¿¿¿¿B (I¿¿¿¿B) is a short half-life protein that is affected by UV-induced translation inhibition and additionally can be a substrate of calpain. This project seeks to elucidate a correlation between calpain, I¿¿¿¿B¿¿¿¿, translation inhibition, and cell viability upon UVB-irradiation, utilizing two cell lines with three key mutants, three inhibitors, flow cytometry, and Western blotting. Results show that calpain does not actively degrade I¿¿¿¿B¿¿¿¿ upon UVB irradiation, but that translation inhibition alone cannot account for decreased I¿¿¿¿B¿¿¿¿ levels.
