The release behavior of the two systems was studied for the release of red dye from cast poly(ethylene oxides) (PEO) and release of Tetracycline hydrochloride from poly(vinyl alcohol) (PVA).Four different molecular weights of polyethylene oxide 200kd, 300kd, 600kd, and 900kd were used to evaluate the release as a function of molecular weight of PEO. The prepared film samples were then compared based on their release kinetics and swelling behaviors. The swelling and dissolution characteristics of four polyethylene oxide grades were studied as a function of time, by monitoring the weight change of film upon exposure to water, and the rates of water penetration, while measuring the release of dye using Uv-vis spectroscopy.Two thicknesses of PEO film were 85μm & 145μm for the evaluation of PEO release rate. The prepared films exhibit different release mechanisms as molecular weight changes. The release from thin-film polyethylene oxide is primarily related to the dissolution rate of the polymer. PEO thin films showed no visible gel-layer upon exposure to water. The total release time increases as molecular weight increased, and the initial burst release of dye decreases as molecular weight increase.Linear release profiles from 200kd PEO thin samples were calculated, while two linear regions for the first two-thirds of release were observed from thinker 200kd samples and all three remaining PEO grades (300kd, 600kd, and 900kd) at the two thicknesses. The release of red dye from 85μm and 145μm samples for 200kd and 300kd have shown dependence on sample thickness, as thinner samples seem to have higher release rate compared to the thicker samples from the same molecular weight PEO. However, higher molecular weight (600kd and 900kd) PEOs, showed less dependence on thickness. The release rate from thinner samples tends to have a close release rate to those from the thicker samples for the first part of the release. The dependence of release rate on molecular weight suggests that the rate-limiting step is polymer dissolution and there is a minimal effect of the gel-layer on the release from PEO thin film.Poly(vinyl alcohol) films were cast with 1 & 2 wt% concentration of TCH. The crystallinity, release rate, and drug activity were calculated. Two thicknesses were considered (138μm & 257μm). The release from the physically cross-linked PVA showed a correlation to drug load as total release time increases as a function of drug concentration. The release rate from the two tested thickness showed similar profiles until the thinner sample releases most of its content then starts to diverge. The reported type of release reflected a swelling control release type, which is expected to a decrease in release rate as a function of time since the drug molecules closer to the surface diffuse out first, and as the release continuous the drug molecules with a longer distance to travel will diffuse slowly.
