ABC-DLBCL exhibits worse outcome following standard therapy. The pathogenesis of ABC-DLBCL is due to constitutive activation of NF-κB signaling pathway which lead to activation of numerous of processes like cell proliferation and survival. Recently, findings indicate ezrin inhibitors (Ez-Is), NSC305787 and NSC668394, were found to disrupt BCR signaling and decrease pro-survival signaling through induction of apoptosis. To gain more insights into the mechanism of action of Ez-Is in ABC-DLBCL, we performed a multiomics analysis of RNA microarray, protein and phosphoprotein mass spectrometry (MS) from ABC-DLBCL cell lines treated with Ez-I. Our results revealed Ez-I affect many genes, protein and phosphoproteins. The pathway enrichment of them showed pathways mainly related to BCR signaling, Rho GTPase signaling and apoptosis are affected.
