Biology Human leukocyte antigen A (HLA) antigens are histocompatibility antigens found on all nucleated cells. HLA antigens and neutrophil(PMN)-specific antigens are found on PMNs. Fragments of HLA antigens are found on mature erythrocytes (RBC). Antibodies directed against foreign antigens occur as a result of stimulation through exposure resulting from transfusion, pregnancy, or tissue graft. The purpose of this investigation was to study influences on the production of leukocyte (WBC) antibodies. A new method was used to harvest lymphocytes and PMNs from a single blood sample. The WBC-rich portion of the blood was layered over a density gradient column containing two solutions of ficoll-hypaque. After centrifugation lymphocytes and PMNs, 95-99% viable, were collected and used. Blood was obtained from three categories of transfused patients: chronic renal disease, immunosuppressed and clinically miscellaneous. Sequentially collected serum samples showed both subject and group variation in the presence of antibody. The variations may be caused by the use of immunosuppressant drugs, the use of WBC-poor blood, and/or the individual's natural ability to produce antibody . Comparison of antibody activity showed that most individuals produced multiple antibodies rather than only one antibody. The only difference seen in antibody response between the chronic renal disease and the immunosuppressed groups was in activity against RBCs. Antibodies directed against lymphocyte antigens were developed only in conjunction with those directed against PMNs . Data suggests that a relationship exists between high-titered, low-avidity (HTLA) antibodies and WBC antigens. These results suggest that in immunologically competent patients the use of component therapy is much more important than in immunosuppressed patients.
