Biology Various cyanobacterial inhibitors and a cyanobacteriocidal toxin produced by Bdellovibrio bacteriovorus (BdAS) were compared for their effect on Phormidium luridum, with regard to morphology, ultrastructure, photosynthesis, respiration, in vitro ferricyanide reduction and electron transport cytochemistry. Electron microscopy reveals a similar cellular response to different cyanobacterial inhibitors, i.e., dichloro-methylurea (DCMU); potassium cyanide (KCN); and BdAS, i.e., formation of intrathylakoidal spaces. At 2000 fc, whole cells of P. luridum produced 405.4 ul 02/mg chl a/hr while fresh spheroplasts produced 326.7 ul 02mg chl a/hr, an efficiency of 80%. The respiration rate of spheroplasts was higher than whole cells.The rate of ferricyanide reduction by whole cells of P. luridum was 126.6 uM/mg chl a/hr. Higher ferricyanide reduction rates were observed in polymyxin B-treated cells (451.2 uM/mg chl a/hr) and spheroplasts (331.2 uM/mg chl a/hr). The ferricyanide reduction rate was stimulated by 0-phenanthroline and BdAS and decreased by DCMU, KCN, ouabain and rotenone.Copper ferrocyanide electron cytochemistry was non-specific except in cells treated with sodium lauryl sulfate. Deposition of 3, 3-diaminobenzidine (DAB) was seen in the intrathylakoidal space. Cytochemical localization, using tetranitroblue tetrazolium (TNBT) was slightly inhibited by BdAS and severely inhibited by ouabain and polymyxin B. Cytochemical results showed that the photosynthetic lamellae of P. luridum also functioned as reactive sites for respiration. The intrathylakoidal space of P. luridum possessed cytochrome c oxidase activity similar to that localized in intracristal spaces of eukaryotic cells.
