Biology The cell membrane phospholipids of Staphylococcus aureus, strains U9W and a Mitomycin C resistant isolate, NR6, were examined in an attempt to correlate antibiotic resistance with phospholipid content. The 32p phospholipid fractions, phosphatidylglycerol (PG), phosphatidylethanolamine (PE) and cardiolipin (CL) were isolated and identified by a combination of thin layer and paper chromatography techniques. Phosphatidylglycerol was found to cornpose the major fraction of the extractable lipids. No differences were detected between strains with regard to the major qlycerol phosphatides however the results indicated the possibility of differences in the concentration of the minor PE and PG fractions. Although the concentration of CL appeared to be the same on both strains, results also indicated that there are differences in the fatty acid substitutions. Examination of the cell wall amino acids, amino sugars and phosphates revealed no quantitative or qualitative differences between the strains. Aspartic acid however did appear in a concentration too great to be attributed to contamination. The N-acetylmuramic acid to N-acetylglucosamine ratio was calculated to be 2.43 and 2.73 per sample for the parent and mutant respectively.
