Material and Methods: An MTT assay was performed on the extracts of the tested materials. The culture medium alone was used as a negative control and copper strip extracts were used as a positive control. Results: A one-way Analysis of Variance (ANOVA) showed a difference between all of the groups in all of the cell lines and Tukey?s post-hoc test revealed no significant differences between the stainless steel orthodontic bands and the negative control group. There was a significant difference between the negative control group and the stainless steel orthodontic bands with silver solder joints in the HaCaT and MRC-5 cell lines. A great variability in the percentages of cell viability was observed. According to these results, the stainless steel orthodontic bands did not induce cytotoxic effects on any of the cell lines. When associated with silver solder, the orthodontic bands decreased the cell viability in the HaCaT, HGF, MRC-5, and Vero cell lines, but there was only a statistical significance in the HaCaT and MRC-5 cell lines. Conclusions: More than one type of cell line must be tested in a cytotoxicity assay due to the outcome variability from one cell line to another. === Introdu??o: Os objetivos deste estudo foram avaliar, in vitro, a citotoxicidade dos an?is ortod?nticos com e sem soldagem ? prata e comparar a viabilidade celular nas linhagens FGH, HaCaT, MRC-5 e Vero ap?s a exposi??o aos extratos dos an?is ortod?nticos. Materiais e M?todos: Foi realizado o ensaio de redu??o de MTT ap?s a exposi??o das culturas celulares aos extratos dos materiais testados. Foram utilizados como controles negativo e positivo, respectivamente, meio de cultura DMEM e extratos de tira de cobre. Resultados: A an?lise de vari?ncia de uma via (ANOVA) mostrou que houve diferen?a estatisticamente significativa entre os grupos em todas as linhagens avaliadas. O teste de compara??es m?ltiplas de Tukey detectou aus?ncia de diferen?a significativa entre os grupos controle negativo e an?is ortod?nticos de a?o inoxid?vel. Houve diferen?a estatisticamente significativa entre os grupos controle negativo e an?is ortod?nticos de a?o inoxid?vel com soldagem ? prata nas linhagens celulares HaCaT e MRC-5. Observou-se grande variabilidade nas percentagens de viabilidade celular entre as linhagens. Conclus?es: Ao avaliar a citotoxicidade de an?is ortod?nticos com e sem soldagem ? prata, existe variabilidade nos resultados quando linhagens diferentes s?o comparadas. An?is ortod?nticos de a?o inoxid?vel n?o induziram efeitos citot?xicos em nenhuma das linhagens testadas. Quando associados ? solda de prata, os extratos dos an?is ortod?nticos reduziram a viabilidade celular nas linhagens FGH, HaCaT, MRC-5 e Vero, embora essa redu??o tenha ocorrido de forma estatisticamente significativa apenas nas linhagens HaCaT e MRC-5.
