O interesse pela obtenção de chalconas e suas respectivas 2,4-dinitrofenilidrazonas se deve às inúmeras propriedades farmacológicas descritas para estas classes de compostos, a saber: analgésica, antiinflamatória, anticâncer, antioxidante, antimalarial, antifúngicas, bactericidas, tripanocidas, leishmanicidas. Neste presente trabalho foram obtidos vinte e seis análogos de chalconas através de reações de condensação aldólica de Claisen-Schmidt. Para a obtenção das chalconas de cadeia longa foram, inicialmente, sintetizados os aldeídos de cadeia longa via reação de O-alquilação. Para isso foram utilizados o 4-hidroxibenzaldeído e 3-hidroxibenzaldeído em reação com os haletos de alquila e 1-clorohexano, 1-clorooctano, 1-bromononano, 1-clorodecano, 1-clorododecano e 1-clorotetradecano. Os aldeídos alquilados e dezoito aldeídos comerciais foram condensados à acetofenona para a obtenção das chalconas que foram, então, condensadas à 2,4-dinitrofenilidrazina para a formação de vinte e cinco 2,4-dinitrofenilidrazonas. Dentre as vinte e cinco hidrazonas aqui sintetizadas, doze foram obtidas na forma de uma mistura de isômeros E/Z e treze hidrazonas foram obtidas como um único isômero. Dentre aquelas doze hidrazonas obtidas como mistura E/Z, quatro foram isoladas como um único isômero por CCD preparativa. As chalconas inéditas e hidrazonas sintetizadas estão sendo avaliadas quanto a suas atividades antiinflamatória, antioxidante e antimalarial. === This study describes the synthesis of chalcone derivatives and their 2,4- dinitrophenylhydrazones, resulting in twenty-three novel compounds being: four analogues of chalcones and nineteen 2,4-dinitrophenylhydrazones. The interest in obtaining chalcones and their 2,4-dinitrophenylhydrazones analogues has grown because of numerous pharmacological properties described in the literature that these compounds have, namely: analgesic, anti-inflammatory, anticancer, antioxidant, antimalarial, antifungal, bactericides, trypanocidal and antileishmanial activity. In the present study were obtained twenty six chalcones analogues via reactions of aldolic condensation of Claisen-Schmidt. For the obtaining of long chain chalcones were initially synthesized long chain aldehydes by Oalkylation reaction. For this, were used the 4-hydroxybenzaldehyde and 3- hydroxybenzaldehyde reacted with alkyl halides 1-chlorohexane, 1-chlorooctane, 1- bromononane, 1-chlorodecane, 1-chlorododecane and 1-chlorotetradecane. The alkyl aldehydes and eighteen commercial aldehydes were condensed to acetophenone to obtain chalcones which were then condensed with 2,4-dinitrophenylhydrazine to form 2,4- dinitrophenylhydrazones. Of the twenty-five hydrazones summarized here, twelve were obtained as a mixture of E/Z isomers and thirteen hydrazones was obtained as a single isomer. Among those twelve hydrazones obtained as E/Z mixture, four compounds were isolated as a single isomer by preparative TLC. The synthesized chalcones and hydrazones are being tested for anti-inflammatory, antioxidant and antimalarial.
