Summary: | 【Objective】The research aimed at identifying the promoter activities of Nicotiana sylvestris Calcineurin B-like Protein family gene NsylCBL10 in the developmental processes and light-responsive processes of tobacco plants. 【Methods】The promoter of NsylCBL10 was cloned and its cis-acting regulatory elements were analyzed. A fusion vector NsylCBL10pro::GUS was constructed and transformed into Nicotiana tabacum. Besides, GUS staining assay was conducted to analyze the tissue-specificity and light-responsive characteristics of NsylCBL10 promoter using NsylCBL10pro::GUS transgenic plants. 【Results】NsylCBL10 promoter was predicted to contain various cis-acting regulatory elements, including tissue-specific elements, light-responsive elements, phytohormone responsive elements and stress-responsive elements. The activity of NsylCBL10 promoter was mainly detected in cotyledon node-root transition section and root tips during germination stage, while the active region expanded to cotyledons during cotyledon stage. The activity was detected in leaves during cross-seedling stage, as well as in style, mature anther, root tips and lateral root base of adult plants. Additionally, it was found that the activity of NsylCBL10 promoter was down-regulated under light condition while up-regulated in dark. 【Conclusions】NsylCBL10 promoter in tobacco plants was of a dynamic activity, which is high in tissues with strong-meristematic ability and floral organs of tobacco plants. Besides, the promoter activity of NsylCBL10 in hypocotyl is greatly regulated by the light signal. |
